An amperometric biosensor based on laccase, from Trametes versicolor (LTV), was developed and optimized for monitoring the phenolic compounds content in tea infusions. The fungal enzyme was immobilized by entrapment within polyvinyl alcohol photopolymer PVA-AWP (azide-unit pendant water-soluble photopolymer) onto disposable graphite screen-printed electrodes (SPE). Sensitivity optimization in terms of pH, temperature and applied potential was carried out. The linear range, detection limit, operational and storage stabilities were also determined. The laccase biosensor (LTV-SPE) was calibrated for o-, m- and p-diphenol as well as caffeic acid. The highest response was found at 0.1M acetate buffer pH 4.7, though it must be added the good reproducibility and operational stability were also obtained. The useful lifetime of the biosensor is estimated to be greater than 6 months. LTV-SPE was used for the determination of the equivalent phenol content (EPC) in tea infusions by the direct addition into the electrochemical cell: the results were compared with those from the Folin-Ciocalteu spectrophotometric method. The amperometric detection exhibits some interesting advantages such as high simplicity, minimal sample preparation and shorter response time. A stable and sensitive amperometric response was obtained toward standard diphenolic compounds and herbal infusions. These biosensors are useful for easy and fast monitoring of EPC that can be related to the antioxidant capacity of natural extracts.

The Sonogel electrode was immersed in hemin solution to obtain the hemin-Sonogel-modified electrode by cyclic voltammetry (CV). It was applied to detect superoxide radicals produced by xanthine oxidase catalyzed hypoxanthine oxidation. The characterization of hemin-Sonogel was modified by impedance and CV.

This works presents the development of a detoxification system based on bacterial phosphotriesterase (PTE) for the degradation of organophosphate (OP) insecticides in water. PTE was immobilised on an activated agarose gel via covalent coupling. Two different OPs were studied, chlorpyrifos and chlorfenvinfos, due to their importance in the field of water policy. The efficiency of insecticide degradation was controlled using a highly sensitive biosensor allowing the detection of OP concentration as low as 0.004mugL(-1). Under optimum conditions, it was shown that a column incorporating 500IU of PTE was suitable for the detoxification of solutions containing either isolated pesticides or pesticides mixtures, even at concentrations higher than authorized limits. Finally, the method was shown to be adapted to the decontamination of real samples of pesticides with concentrations up to 20mugL(-1).

This paper describes the characterization of the gold-catalyzed deposition of silver on graphite screen-printed electrodes (SPEs) using electrochemical impedance spectroscopy (EIS) and the application of this approach to the development of impedimetric immunosensors. After applying -0.1 V for 45 s, the amount of electrodeposited silver quantitatively changes the magnitude of two elements of the electrical equivalent circuit: the interface capacitance, Ci, and the charge-transfer resistance, R(CT). Better correlations have been found when considering the R(CT) since this parameter is almost exclusively dependent on the amount of deposited silver under these experimental conditions. This approach has been successfully applied to the development of an impedimetric immunosensor for aflatoxin M(1). The R(CT) magnitude shows good correlation with the amount of gold immobilized on the electrode surface after a competitive assay and thus, with the toxin concentration. This approach has been found sensitive in a wide range of concentrations, from 15 to 1000 free-AFM(1) ppt with a limit of detection of 12 ppt.

Amperometric acetylcholinesterase (AChE) biosensors have been developed to resolve mixtures of chlorpyrifos oxon (CPO) and chlorfenvinfos (CFV) pesticides. Three different biosensors were built using the wild type from electric eel (EE), the genetically modified Drosophila melanogaster AChE B394 and B394 co-immobilized with a phosphotriesterase (PTE). Artificial Neural Networks (ANNs) were used to model the combined response of the two pesticides. Specifically two different ANNs were constructed. The first one was used to model the combined response of B394+PTE and EE biosensors and was applied when the concentration of CPO was high and the other, modelling the combined response of B394+PTE and B394 biosensors, was applied with low concentrations of CPO. In both cases, good prediction ability was obtained with correlation coefficients better than 0.986 when the obtained values were compared with those expected for a set of six external test samples not used for training.

This paper describes the characterization and optimization of an amperometric cytochrome c (cyt c)-based sensor for the determination of the antioxidant capacity of pure substances and natural samples. The cyt c and the xanthine oxidase (XOD) enzyme were co-immobilized on the electrode using the combination of several long-chain thiols. The self-assembled monolayer (SAM) was optimized in terms of composition and ratio between thiols. The immobilization protocol for both cyt c and XOD and the SAM formation time were evaluated through electrochemical methods, such as cyclic voltammetry (CV), square wave voltammetry (SWV), chronoamperometry (CA) and impedance spectroscopy (IS). Finally, the biosensor was applied to the determination of the antioxidant capacity of pure alliin and two compounds extracted from garlic bulbs.

This paper describes the development of an amperometric cytochrome c (cyt c)-based biosensor and its later application to the quantification of the scavenging capacity of antioxidants. The enzymatic biosensor was constructed by covalently co-immobilizing both cyt c and XOD on a mercaptoundecanol/mercaptoundecanoic acid (MU/MUA) mixed self-assembled monolayer (SAM)-modified screen-printed gold electrode. The applicability of this method was shown by analyzing the antioxidant capacity of pure substances, such as ascorbic acid and Trolox, and natural sources of antioxidants, particularly 5 orange juices.

This article gives an overview of the electrochemical biosensors that incorporate genetically modified enzymes. Firstly, the improvements on the sensitivity and selectivity of biosensors that integrate mutated enzymes are summarised. Next, new trends focused on the oriented immobilisation of mutated enzymes through specific functional groups located at their surface are reviewed. Finally, the effect of enzyme mutations on the electron transfer distance and kinetics of electrochemical biosensors is described.

This work shows the possibility of combining the high sensitivity of genetically-modified Drosophila melanogaster acetylcholinesterase (B394) with the ability of phosphotriesterase (PTE) to hydrolyse organophosphate compounds, in the aim of developing a biosensor selective to two insecticides of interest: chlorpyrifos and chlorfenvinfos. The studies clearly demonstrate that chlorfenvinfos is a substrate that acts as competitive inhibitor of PTE, therefore preventing the efficient hydrolysis of other pesticides, including chlorpyrifos. A bi-enzymatic sensor was designed by immobilizing both B394 and PTE in a polyvinylalcohol matrix. The sensor was shown to be able to discriminate between chlorpyrifos and chlorfenvinfos inhibitions.

In this paper, the inhibition effect of aflatoxin B1 on different species of cholinesterases was investigated to unravel action mechanism. The inhibition curves of several cholinesterase mutants (obtained by spectrophotometric measurements of enzyme activity, pS curves) were analyzed. They showed that this toxin reversibly inhibits cholinesterases by binding to a peripheral site located at the entrance of the active site gorge without entering inside the site. Electric eel enzyme revealed the highest inhibition extent with a binding constant estimated to 0.35 microM. This binding prevents the entrance of substrate en route to the catalytic site and also decreases chemical steps of the reaction at the catalytic site: acetylation is reduced to the half and deacetylation is reduced to the third. Electric eel acetylcholinesterase was used to settle an amperometric biosensor. The best detection was obtained by using 0.3 mU enzyme on the electrode and 0.5mM ATCh in the solution. The limit of detection was 3 microM corresponding to 20% inhibition.

This article describes the different types of marine toxins and their toxic effects, and reviews the bio/analytical techniques for their detection, putting special emphasis to biosensors. Important health concerns have recently appeared around shellfish (diarrheic, paralytic, amnesic, neurologic and azaspiracid) and fish (ciguatera and puffer) poisonings produced by different types of phycotoxins, making evident the urgent necessity of counting on appropriate detection technologies. With this purpose, several analysis methods (bioassays, chromatographic techniques, immunoassays and enzyme inhibition-based assays) have been developed. However, easy-to-use, fast and low-cost devices, able to deal with complicated matrices, are still required. Biosensors offer themselves as promising biotools, alternative and/or complementary to conventional analysis techniques, for fast, simple, cheap and reliable toxicity screening. Nevertheless, despite the wide range of seafood toxins and the already rooted biosensing systems, the literature on biosensors for phycotoxins is scarce. This article discusses the existing biosensor-based strategies and their advantages and limitations. Finally, the article gives a general overlook about the regulation toxin levels and monitoring programmes currently established around the world concerning seafood safety.

An electrochemical biosensor for the detection of microcystin has been developed based on the inhibition of the protein phosphatase 2A (PP2A) by this cyanobacterial toxin. The enzyme has been immobilised by entrapment using a poly(vinyl alcohol) azide-unit pendant water-soluble photopolymer (PVA-AWP). Electrode supports and immobilisation conditions have been optimised by colorimetric assays, the highest immobilisation yields being obtained with screen-printed graphite electrodes and the 1:2 PP2A:PVA ratio. Catechyl monophosphate (CMP), alpha-naphthyl phosphate (alpha-NP) and 4-methylumbelliferyl phosphate (4-MUP) have been used as phosphorylated substrates to monitor the protein phosphatase activity by electrochemical methods, the former providing the highest chronoamperometric currents at appropriate working potentials (+450mV versus Ag/AgCl). Incubation with standard microcystin solutions has demonstrated the inhibition of the immobilised enzyme, proportional to the toxin concentration. The standard inhibition curve has provided a 50% inhibition coefficient (IC(50)) of 83mugL(-1), a limit of detection (LOD; 35% inhibition) of 37mugL(-1), and 100% inhibition at about 1000mugL(-1). Real samples of cyanobacterial blooms from the Tarn River (Midi-Pyrénées, France) have been analysed using the developed amperometric biosensor and the toxin contents have been compared to those obtained by a conventional colorimetric protein phosphatase inhibition (PPI) assay and high-performance liquid chromatography (HPLC). The results clearly justify the use of the developed amperometric biosensor as screening method for microcystin detection.

A novel label-free electrochemical impedimetric immunosensor for sensitive detection of ochratoxin A (OTA) was reported. A two-step reaction protocol was elaborated to modify the gold electrode. The electrode was first derivatized by electrochemical reduction of in situ generated 4-carboxyphenyl diazonium salt (4-CPDS) in acidic aqueous solution yielded stable 4-carboxyphenyl (4-CP) monolayer. The ochratoxin A antibody was then immobilized making use of the carbodiimide chemistry. The steps of the immunosensor elaboration and the immunochemical reaction between ochratoxin A and the surface-bound antibody were interrogated using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The impedance change, due to the specific immuno-interaction at the immunosensor surface was utilized to detect ochratoxin A. The increase in electron-transfer resistance (DeltaR(et)) values was linearly proportional to the concentration of OTA in the range of 1-20ngmL(-1), with a detection limit of 0.5ngmL(-1).

Three strategies have been compared to produce screen-printed amperometric detectors for NADH: mixing Meldola Blue (MB) in the screen-printing ink, incorporation of MB-Reinecke salt (MBRS) in the graphite ink and electrodeposition of films of MB-derived polymer (poly (MB)) on electrode surface. Following modification of graphite electrodes the mediators displayed values of the formal potential E degrees ' from -0.129 to -0.160 V vs. Ag/AgCl and pK(a)s of 5.09-6.02. A second redox couple with E degrees '=-0.450 V vs. Ag/AgCl was observed in cyclic voltammetry experiments with poly (MB) sensors or with old electrodes obtained according to the other two strategies. Electropolymerisation of MB allowed to achieve the best operational stability and best detection limit, 2x10(-6) M, for amperometric detection of NADH, while the most extended linear range, 1x10(-5)-7.5x10(-4) M, corresponds to sensors with MBRS. MB and MBRS electrodes were compared with a similar NADH detector produced by Gwent Electronic Materials, England. Several characteristics of the modified-electrodes induced by the fabrication by screen-printing were also highlighted.

A screen-printed biosensor for the detection of pesticides in water miscible organic solvents is described based on the use of p-aminophenyl acetate as acetylcholinesterase substrate. The oxidation of p-aminophenol, product of the enzymatic reaction was monitored at 100 mV vs. Ag/AgCl screen-printed reference electrode. Miscible organic solvents as ethanol and acetonitrile were tested. The acetylcholinesterase (AChE) was immobilised on a screen-printed electrode surface by entrapment in a PVA-SbQ polymer and the catalytic activity of immobilised AChE was studied in the presence of different percentages of organic solvents in buffer solution. The sensor shows good characteristics when experiments were performed in concentrations of organic solvents below 10%. No significant differences were observed when working with 1 and 5% acetonitrile in the reaction media. Detection limits as low as 1.91x10(-8) M paraoxon and 1.24x10(-9) M chlorpyrifos ethyl oxon were obtained when experiments are carried out in 5% acetonitrile.

Screen-printed electrodes coated with the nafion layer have been investigated for cholinesterase biosensor design. The butyrylcholinesterase (ChE) from horse serum was immobilised onto the nafion layer by cross-linking with glutaraldehyde vapours. The biosensors obtained showed better long-term stability and lower working potential in comparison to those obtained with no nafion coating. The sensitivity of a biosensor toward organophosphate pesticides is not affected by the nafion coating. The detection limits were found to be 3.5x10(-7) M for trichlorfon and 1.5x10(-7) M for coumaphos.

The exponential development of biosensors as powerful analytical tools in the last four decades mainly relies on the high sensitivity and selectivity offered when detecting the target analyte. The transducer and the biological receptor are the bases of the biosensor development. Nevertheless, the bioreceptor immobilisation is also important, playing a key role in the retention of the biological activity, and thus affecting the sensitivity. Parameters such as shelf-life and surface regeneration also depend on the biomolecule immobilisation. Researchers are focusing their efforts towards random and oriented immobilisation procedures. Adsorption, entrapment, cross-linking and electrostatic interactions provide randomly immobilised biomolecules, sometimes partially hindering their biological activity. Covalent binding and affinity interactions may enable oriented biomolecule immobilisations, providing controlled, reproducible and highly active modified surfaces. This paper reviews the main immobilisation strategies used in the biosensors development, putting special emphasis on our contribution to mild and oriented immobilisation techniques.

Two new amperometric biosensors based on immobilization of acetylcholinesterase on a sonogel-carbon electrode for detection of organophosphorous compounds are proposed. The electrodes were prepared applying high-energy ultrasounds directly to the precursors. The first biosensor was obtained by simple entrapping acetylcholinesterase in Al(2)O(3) sol-gel matrix on the sonogel-carbon. The second biosensor was produced in a sandwich configuration. Its preparation involved adsorption of the enzyme and modification via a polymeric membrane such as polyethylene glycol and the ion-exchanger Nafion. The optimal enzyme loading was found to be 0.7 mIU. Both biosensors showed optimal activity in 0.2 M phosphate buffer, pH 7.0, at an operating potential of 210 mV. The detection limit achieved for chlorpyriphos-ethyl-oxon was 2.5x10(-10)M at a 10-min incubation time.

Five different clones of antibodies developed against the aflatoxin M(1) were investigated by using the classical indirect and direct competitive Enzyme-Linked Immunosorbent Assay (ELISA) formats, and also the direct competitive ELISA based on the use of the superparamagnetic nanoparticles. The purpose of this study was to assess if not so friendly time classical ELISA procedures can be further improved, by reducing the coating, blocking and competition time. Here we showed that a complete dc-ELISA (coating, blocking and competition step) based on the use of superparamagnetic nanoparticles can be performed in basically 40 min, if coating step (20 min) should be taken into account. Moreover, the standard analytical characteristics of the proposed method fulfil the requirements for detecting AFM(1) in milk, in a wide linear working range (4-250 ng/L). The IC(50) value is 15 ng/L. The matrix effect and the recovery rate were assessed, using the European Reference Material (BD282, zero level of AFM(1)), showing an excellent percentage of recovery, close to 100%.

Electrochemical immunosensors based on a competitive indirect enzyme-linked immunosorbent assay (ciELISA) and an enzymatic recycling system were developed for the detection of okadaic acid (OA). OA-ovalbumin (OA-OVA) conjugate was immobilised on screen-printed electrodes (SPEs) and competition of a newly generated monoclonal antibody (MAb) for free and immobilised OA was subsequently performed. Secondary antibodies labelled with alkaline phosphatase (ALP) or horseradish peroxidase (HRP) were used for signal generation. Experimental parameters were firstly optimised by colorimetric ELISA on microtiter wells and on SPEs. The ELISA system was then tested by amperometry at +300 mV vs. Ag/AgCl (detection of p-aminophenol produced by the reaction of p-aminophenyl phosphate with ALP) or -200 mV vs. Ag/AgCl (detection of 5-methyl-phenazinium methyl sulfate, redox mediator in the HRP bioelectrocatalysis). The limits of detection (LODs) with standard solutions were 1.07 and 1.98 microgL(-1) when using ALP and HRP labels, respectively. An electrochemical signal amplification system based on diaphorase (DI) recycling was integrated into the ALP-based immunosensor, decreasing the LOD to 0.03 microgL(-1) and enlarging the working range by two orders of magnitude. Preliminary results with mussel and oyster extracts were obtained and compared with the colorimetric immunoassay, the colorimetric protein phosphatase inhibition assay (PPIA) and LC-MS/MS.
